载多西他赛温敏类弹性蛋白多肽自组装微粒的制备

摘要  目的:制备温敏类弹性蛋白多肽(ELPs)自组装微粒,并考察其温敏特性及对模型药物多西他赛的包载效能。方法:将ELPs冻干粉重新分散于冷的NaCl磷酸盐缓冲液中,利用配备电热温控装置的紫外-可见分光光度计,在波长350 nm处测定ELPs溶液的光密度,升降温速率为1 ℃/min,计算相变温度。向ELPs溶液中加入多西他赛50%乙醇水溶液,混合完全,相变温度下孵育30 min即得载药微粒,保温离心测定包封率并考察体外释放情况。结果:相变温度下自组装形成的载药微粒外观圆整,粒径约为1 μm,包封率可达15.12%。结论:ELPs具有温敏特性,且可实现对多西他赛的包载。
