摘要  目的:探讨三氧化二砷(arsenic trioxide,As2O3)对人卵巢癌细胞迁移、侵袭的影响及其相关分子机制。方法:分别通过细胞划痕、Transwell小室及预包被Matrigel胶的Transwell小室(侵袭小室)方法,观察As2O3对人卵巢癌细胞SKOV3迁移、侵袭能力的影响。蛋白质印迹法检测As2O3处理不同时间后肿瘤转移相关的蛋白,包括Rho相关蛋白激酶(Rho related protein kinase,ROCK)及基质金属蛋白酶家族(matrix metalloproteinase family,MMPs)蛋白的表达。结果:2 μmol/L As2O3即可显著抑制SKOV3细胞的迁移及侵袭。蛋白质印迹法检测发现,As2O3可显著抑制ROCK1、ROCK2及 MMP-9的表达,但对 MMP-2表达无明显影响。结论:As2O3可明显抑制人卵巢癌细胞SKOV3的迁移、侵袭能力,这可能与其抑制ROCK1、ROCK2以及 MMP-9的表达有关。
